Hello Everyone, I need to know about the difference of fate of suicide plasmid in fungi and in bacteria.. As I know, in bacteria, suicide plasmid deletes a gene and is removed in next generations, is it same with plasmids in fungi or it replicates in the next generations..
Secondly, can anyone help me with gene sequences of the markers used in gene knockout in fungi,

Normally suicide plasmids are either non-replicating or conditional replicating so they would be lost in subsequent generations. With the right markers however you can select against some plasmids and ensure they are lost even if they are replicating, this is most commonly done with yeast experiments.
You should be able to find the sequences you are looking for in Genbank.
I know that ncbi can help meto get the sequences, main thing i want to know is the names of selection markers used commonly or specifically in fungi gene manipulation, thank you..
There are lots of different fungi and different markers work best in certain strains. Why don't you review the literature for the organism that you are working with to determine the best.
