I am trying to see the effect of a drug on cell viability. The drug was dissolved in sodium phosphate buffer (50mM). In control cells where no drug was added but the buffer was added, a lot of cell death was seen. What is the reason for that?
Assuming that (i) a small volume (as compared with cell culture medium) of the sterile buffer was added to cells; (ii) the pH of the buffer was adjusted before the addition; and (iii) cells were incubated for a short time (not exceeding 12-16 h) with the buffer, the most likely reason would be that the drug treatment protected cells from cell death under culture conditions used. More culture conditions details (see above) could help identify the scientific reason.
