If expanding the ring structure from 5 member to 6 or 7 , then what is the effect of ring size on the MDR, bioavailability and cytotoxicity in general.
Ring size is generally correlated with lipophilicity. An increase in the ring size from 5 to 7 will increase the lipophilicity of the molecule.
If the drug is hydrophilic and you increase the ring size contained in the drug, the bioavailability will be increased and vise versa.
In the above mentioned case if your drug is hydrophilic anti-cancer agent and you increase the ring contained in the molecule the cytotoxicity will be increased (positive result).
There is NO correlation between MDR and ring size.
