I want to do protein estimation by Lowry's Procedure. For standard, i have to prepare standard curve with bovine serum albumin (100 microgram /ml) . I have to prepare a standard curve upto 150 microgram BSA concentration.
Should i dissolve ist BSA 10 mg in 100 ml distilled water. Out of this solution, i have to take different concentration (0.1,0.2,0.3 to 1.5 ml) for preparation of standard curve
Hi Priya, the easiest way to do this is to make a stock solution (BSA 0.1 mg/ml) and do a serial dilution of the stock into water.
For example, you have 1 ml of 0.1mg/ml BSA. Line up eppendorf tubes with 100 ul water. Pipet 100 ul of the stock, place into the next tube of water, pipette up and down, transfer the next 100 ul into the next tube and repeat.
Then you will have BSA stock solutions of 100ug/ml, 50 ug/ml, 25 ug/ml, etc.
Weigh 100 mg BSA and dissolve in 10 mL water to make 10 mg/mL BSA solution as stock. Then prepare solutions of 100 μg/mL by diluting 100 μL BSA (10 mg/mL) with 9.9 mL water.
