i am doing a Osteogenic differentiation for 21 days, but the growth of my mesenchymal stem cells isn't stoped .
i am worry about over growth of them and after that starting the apoptosis procedure and because of that i can't have my cells for 21 day.
i heard that if i remove FBS from my medium the growth of the cells become limited .
is it true?
is it has a bad effect on my cells or my differentiation procedure?
what's your suggestion?

FBS depletion leads to cell detachement and death, unless you supplement medium with gorwth factor likewise FGF, EGF and other one.

Not sure what kind of experiment you are doing and what you want to achieve, but why don't you decrease the initial cell density/well? Maybe they are too packed and a change of cell density will help (?)
What % FBS are you using? You can reduce to 1-2% FBS in media. That will slow the growth.

i'm using 10% FBS in my medium, do you think the growth of cells become lower with lowering FBS.

you could try 2% Horse serum that normaly is used to block cell growth and promote myogenic differentiation.

i always use bovine serum and i can't use horse serum because we don't have that in our lab.
2% is good but i have problem with promoting myogenic differentiation because i want to stimulate cells for osteogenic differentiation.
do you think 2% just block cell growth and doesn't have any other effect on cells ( like apoptosis, ... ) ?

2% FBS could be too low and induce cell eath, you could try 2 or 5% FBS, HS is much reacher than FBS.
