Dear all
I wish to stain the nucleolus of A549 for confocal microscopy but I don't want a dye which stains the RNA because the protein I am monitoring is RNA-binding, meaning that a RNA-targeting dye may give false positive. Any suggestion on my options? Any advice is appreciated.

To my knowledge, with the exception of RNA-selective stains, there is no way to "stain" a cell for the nucleolus. If your cells are compatible with transient transfection, then you can use a fluorescent protein fusion to Nucleolin or Fibrillarin to visualize the nucleolus.
