Hello everyone, we are trying to isolate DNA from fecal samples without using any kit. Any help regarding the protocol would be much appreciated. Thanks in advance.
I had used the CTAB/PCI method from the below paper. It did not work as I was expecting, but one of my colleagues had fairly good success with this. See if this works for you. I found that if you have fresh fecal samples, then the kits work best.

As suggested by Kunal Arekar, you can use the CTAB method (Corach et al., 1995) as in "Oklander, L. I., Marino, M., Zunino, G. E., & Corach, D. (2004). Preservation and extraction of DNA from feces in howler monkeys (Alouatta caraya). Neotrop Primates, 12(2), 59-63".
Follow the advice from comments above. But do remember that doesn't matter what protocol you use, you might need to do few adjustments based on your sample type. Every sample type is unique in its own way and can have different amount of different inhibitors.
