What is the difference between using southern blot and gene complementation after a knockout gene?

Why did we use complementation inspite of how we examine how many copies of the gene mutated by southern blot?

Hi Ayat:

Depends on what method did you use for gene knockout:

Southern-- to check on 'DNA' level

complementation rescue-- to see if you can rescue those 'knockout' plants (using wild-type to provide mRNA, protein)-- mRNA and protein level
