S9 contains both the cytosol and microsomes, in which the cytosol has soluble proteins (e.g. NAT, GST, SULT) and the microsomes are the membrane proteins (e.g. P450, UGT, FMO). The differential low speed centrifugation should first separate the S9 fraction from the waste pellet, a second high speed centrifugation of the S9 fraction will separate the cytosol from the microsomes, with the latter forming the pellet.
