We have performed an experiment with a chemical in our lab and according to our results, the compound is toxic on MCF-7 but it is not toxic on A549. There is a selective effect on cancer cell lines for this compound. So my question is if anybody knows the difference between A549 and MCF-7 cell lines? It might be expressions, proteins, receptors or anything else.
I think there were big differences between A549 and MCF7 in expressing multi drug protein, there were big role of Multi drug resistance proteins in A549 cell line and drug accommodation. MRP1 (multidrug resistance-associated protein) and LRP (lung resistance-related protein) mainly high in A549 cell. At any cytotoxicity assays it might cause that A549 cells were less sensitive to any treatments than MCF7
There is a lot of differences - first of all MCF-7 is a metastatic tumor in contrast to A549 (which is primary tumor). It means that MCF-7 have more mutations what makes them more agressive and more resistant to treatment. They have different expression of growth factor receptors and anti-apoptotic proteins. Furthermore from my experince there are always differences in treatment outcomes in different cancer cell lines. Do the A549 cells are enlarged, flattened and stop to divide after treatment? Maybe they do not die, but they become senescent - viable but anable to proliferate.

Dear Agnieszka Bojko, A549 cells did not stop dividing after treatment. Their viabilities was same with control wells.
