We are trying to achieve knockout of ADE12 gene in S. cerevisiae strain, but we are a little bit confused on expected color of colony.
SGD states that these cells should acquire red pigment, but in citing article ( Dorfman BZ (1969) ) they had ade1 and ade2 background, that itself is pigmented. Ade12 locus is quite seldom used and I have found some articles with white and some with red yeast colonies.
Has anybody worked with this mutant and has first hand knowledge on ade12 mutant color?
They should become red if they are ade mutants. In order to see the red pigment you should remove adenine from the media/plates.
Thank you Alice, as far as we have worked with ade pathway mutants only ade1 and ade2 become red, upstream and downstream mutants ade4, 6, 5-7, ade16ade17 will keep white color. Hence the confusment with ade12

ade12 is very downstream of ade1 and ade2 in the pathway, these 2 being the only mutants leading to red clones. The ade12 mutant is white.
Agnese Kokina I'm curious if you came to a resolution with this question? From what I can tell, the red color is from the ade1/2 mutant background from the original study, not from ade12 itself. Is that correct?
