I'm currently working on a project to study BiTEs, but I'm struggling with the best way to quantify the amount of BiTE being produced by my cells. Any help would be greatly appreciated. I've thought about ELISAs but those pose the problem of it being bispecific; I feel like I would need a capture antibody to look at one FcSv, while the detection antibody would have to detect the second FcSv. Then there would be the problem of a positive control.
I guess it's possible, but I wasn't sure if there's a better method.
Hi, I am struggling with the same issue - having real issues with the dual analyte ELISA. I am considering trying a competitive ELISA for one of the analytes - see how much the signal decreases after incubation with the bite. it will probably only be semi quantitative. There's also the issue that a lot of bite will be stuck to the secreting T cells, so probably flow assay needed for that. If you come up with any other ideas, please share!!!
