Would an ELISA be a better approach? Panels are useful for narrowing down where to look- is there a better way to prep sample or observe cytokines in mouse nasopharynx?
You can use ELISA method and Flow cytometric analysis (Treg) Gene expression (Quantitative real-time PCR) .All the best.
The sensitivity of the kit is really high and so I don't think doing individual ELISAs would fix your problem. Do you know how much of the cytokines you should expect to see? It could be that not much is produced in your model. If the cytokine levels are low, you would have quite a lot of variability between mice and it is normal. One thing you could try is to see if there is any way you could concentrate the samples that you test (maybe wash/lavage in less volume so the cytokines are not diluted out?).
