F4/80 and Iba1 will work. A really nice stain you could try that is usually primary labeled is Phalloidin. It is a toxin that specifically binds F-actin so you get a great structural stain. Any cell type will be compatible.
Identifying macrophages: General macrophage identification can be achieved by using antibodies against pan-macrophage markers, such as F4/80 or CD68.
Distinguishing macrophage subsets: If you want to differentiate between different macrophage subtypes (e.g., M1 vs. M2), iNOS for M1, Arginase-1 for M2, specific antibodies targeting markers unique to each subset are needed.
Visualizing cellular processes: Techniques like phagocytosis or antigen presentation can be visualized using fluorescent probes specific to the process of interest.
General cellular morphology can be observed by staining with Actin Green or Phalloidin Red.
I want to investigate the role of Shigella's virulence effector proteins in killing mechanism/cell death/apoptosis of macrophages via microscopy. I need guidelines about staining in this regard please. Your guidence would be highly appreciated please.
