You can extract (10g dry powder) it through hydro ethanol (Ethanol: Water respectively in 70:30 ratios). and then fractionate (3g hydroethanolic in 60 ml distilled water and mix with 60 ml hexane. Repeat the entire process thrice) it with hexane, chloroform, ethyl acetate, methanol and aqueous. The hydro ethanol will extract many different bioactive compounds. The nonpolar to polar fractions will separate the different bioactive compounds.
You can extract with successive extraction by soxhlet method .non polar to polar solvent used for this type of extraction . then fatty portion will extract with nonpolar & you will get polar extraction which have high flavonoids & phenolic content or hydroethanolic also best after defatted extraction.
In the hydroethanolic extract a wide range of bioactive constituents can be extracted. The mixture of water and alcohol (ethanol) could be more successful solvent mixture than single solvent in the extraction of polyphenols.Water is efficient solvent in extracting polar molecules while in the case of less polar compounds, greater efficiency could be achieved by using organic solvents or binary solvents. This is due to the fact that different polyphenols have different solubility and varying degrees of polarity. Using a sequence of solvents with different polarities allows for the extraction of a broader range of polyphenols than a single solvent, which may not be able to extract certain types of polyphenols.
