I want to transfect differentiated SH-SY5Y cells with AAV-plasmid DNA. Anyone who performed transfection in the differentiated SH-SY5Y cells, please share your experience.

It is hard to get it transfected with Lipo or usual methods. It may reach 10-15% transfectants with Lipo but does not serve the purpose many a times. Roche Fugene should work as many people claim that it does. We used Mirus TransIT 2020 once and it seems to work much better and its cheaper than Roche. So you could try that. Also confluency and cell health of SHSy is very critical while transfecting. Some people also tried below procedure in the past and was able to achieve 40% transfection efficiency.here is the link. It is hard to transfect the SH-SY5Y cells with Lipo transfection reagent.You can transfect the SH-SY5Y cells with adenoviral vector.You can also use the special serotype-AAVDJ, which can be packaged by your AAV plasmid and the Rep-Cap(DJ) plasmid.
