We are planning to transfect a relatively large luciferase vector into cancer stem cells.
Does anyone know now what would be the best transfection reagent for this purpose?
Would lipofectamine work? Would electroporation or lentivirus based system work better?
I will appreciate any suggestion!

In general, large vectors may create some problems with the intracellular delivery, the electroporation or lentiviral transduction work better.
for tough cells electroporation or lenti typically works better, but Lipofectamine 3000 is definitely worth trying- simple protocol vs two above options, and it is the latest addition to the "classic" lipid based arsenal
For in vitro studies, transduction through lentiviral vectors will be extremely efficient. Electroporation may work, but it can result in excess cytotoxicity. With lipofection, you may want to find primary cell reagents (e.g. https://altogen.com/product/altofect-transfection-reagent/) and that could help with compatibility issues. Larger vectors can also lead to problems with membrane penetration, so using a complex condenser can help you avoid issues there as well.
