As we know that Xylazine/Ketamine administration, leads to acute hyperglycemia and subsequently a decrease levels of plasma insulin in fasted rats and this count as sampling error in experiments...!
Hi Nader Hamedchaman , you don't need to anesthetize wistar rats to take blood samples safely. You can use the tail nick method in awake animals without any issues. You could also use a catheter to always have an access to blood, however it requires surgery and daily care that can be very time consuming.
What do you intend to achieve with the anaesthetic agent??? You can collect blood samples from the experimental animals through the tail vein (with some restriction) and put a single drop on the strip without having to anaesthesize the animals.
I have done many hundreds of central vein catheters in rats and can take blood without waking a sleeping rat. This gives a stress free sample that gives true and accurate blood glucose values. I have done this in streptozotocin diabetic and normal animals with superb results. Anesthetics and even brief restraint to get tail blood, impact stress hormones and elevate blood glucose so your results are not going to truly reflect the conditions you are wanting to assess.
