We performed a study on the intestinal microbiota of rats in six intervention groups (8 rats in each group).
It is suggested that since the rats in each group were the same in terms of type of intervention and storage conditions and had the same genetics, the fecal samples of the rats in each group be pool together (ie we have a total of 6 pooled samples for the 6 intervention groups) and Then perform microbiota tests. I want to use STAMP software for statistical analysis
Given the above description, what is the best statistical method for statistical analysis?
should I use from Whithe's non-parametric t-test (two groups) or chi-square test (two samples) or Another method in STAMP?

Does ALL the data you have relate merely to the 6 intervention groups? In addition, what is the primary objective and research design of your study?

The aim of this study was to evaluate the effect of high protein diet and exercise on the intestinal microbiota composition of obese rats.
For this purpose, we had six intervention groups:
1. High fat diet (obese control 1)
2. High fat diet and exercise
3. High protein diet and exercise
4. High protein diet (alone)
5. Normal diet (obesity control 2: discontinue high-fat diet and replace with standard diet)
6. lean control group with normal diet
After collecting fecal samples from all rats, the fecal samples of the rats in each group pooled together (ie we had 6 pooled samples for the 6 intervention groups) and Then perform microbiota tests. (Investigation of microbiota composition and diversity by 16s rRNA method).
Finally, we want to compare the relative abundance of bacteria between groups at the phylum, family, and genus levels and see how each intervention changes the composition of the microbiota.
