We are planning to develop a fluorimetric method for the quantification of mesalamine and its forced degradation products. In that way, we are trying to optimize the solvent for the measurement of the fluorescent intensity of drug. Mesalamine exhibits fluorescent intensity in different solvents and various buffers (see the values are in the table attached below). Can anyone say which of the solvents they think would be the best?
HPLC grade Methanol will be better because the dye mesalamine is purely soluble in methanol.
Some more information will help. Are all the samples in the table normalized to the same concentration? And what is the absorbance at the excitation wavelength for each sample? And what will the original samples be? Biological fluids or tissues? Lab water or wastewater? The final application details might impact three choices made I'm initial method development.

if you want to measure mesalamine in the real sample it is better to use that solvent in which mesalamine has a longer wavelength emission. because if your compound has a blue emission it may overlap with background emission. in this case Phosphate buffer pH6.8 can be best one. on the other hand your compound in DMSO and DMF solvents show the highest intensity, resulting low detection limit. I think you can use the mixture of solvents. in addition some surfactants can be used to increase the fluorescence intensity of the compound.
Polar solvents: alcohols, acetonitrile. Did you investigate dependence I=f(pH) in different buffers
From your results I found DMF or DMSO solvents give higher fluorescence intensity with your drug. Hence you may use DMF or DMSO solvents for your study.
