I am currently injecting mice at P3-7 with BrdU, I use a 50mg/kg dose (concentration at 5-10mg/ml). I noticed that after intraperitoneal injection of volume between 10-30 ul, the BrdU solution can leak through the injection site. This makes giving an accurate dose difficult. Is there an alternative route (subcutaneous for example) that has been shown equivalent to IP injections, such that systemic levels are comparable, which would not cause a such problem? Ultimately, the mice will be sacrificed and BrdU will be detected in the brain.
I inject BrdU subcutaneously in the dorsal neck fold of young pups.That worked well.
Thank you very much Christian. I tried IP injections but I think its too problematic in the post-natal days, especially if the pups underwent a procedure. Is there a specific dosing regime that you used Christian? And did you look at the brain to see if you see BrdU+ cells?
I used the regular 50 mg/kg bodyweight and I diluted my 10 mg/ml stock solution so that I would inject about 20 µl per pup. BrdU was seen mostly in periventricular regions at P1.
