Hi Experts,

I know that there is no perfect methodology for any experiment. But I am just wondering since I have been doing Flow Cytometry experiments and just curious for the best isolation method for astrocytes, microglia and neurons in frozen brain tissue samples?

Could you let me know the best fluorescent tags for these cell types? I read that GFAP for astrocytes may not maximize my yield for isolating astrocytes since GFAP is expressed more in stimulated cells.

The samples will be used for RNA-seq and proteomics. Is it OK to use brains from perfused animals? I read somewhere that the quality is reduced and may not be useful for RNA-seq preparations. I'm not sure for proteomics though.
