1. What is the best possible way to assemble a cell before electrochemical measurements? Coin cell or other ways?
2. What is air-tight HS-Flat test cell?

There are various ways common in literature, such as coin cells assembly or a beaker-cell, where one may use glassy carbon, graphite, Ni-foam, etc as a working electrode.
Personally, I prefer the latter as you have flexibility with the choice of working electrode and reference electrode if you choose a 3-electrode set-up. I also find the assemby quicker so it is easier to test many materials.

for three electrode half cell measurement simple beaker cell is pretty good. But the the working electrode should have proper density for getting good result. for three electrode measurement best universal method for electrode preparation is making a dough using PTFE dispersion as binder.
For two electrode full cell measurement, the pressure between is very crucial for getting good performance. If the electrodes are not tightly held to each other (with a separator in between), then there will higher leakage resistance (or charge transfer resistance).
HS flat test cell is a proprietary test cell manufactured by Hohsen corp. japan and it is very popular for testing electrode materials for supercapacitor or battery.
Coin cell is also good but not the best, because the pressure inside the coin cell is not enough for supercapacitor. Still if you want to use coin cell then its better to use compact one such as CR2025.
