I would like to run an invitro drug release experiment for chitosan nanoparticles (600nm) loaded with herbal extract using dialysis membrane method. However since its not easy to ascertain the molecular weight of the herbal extract because its mixture of different compounds. therefore I am not sure which is the best MWCO for dialysis membrane that I can use. I used 5Kda but nothing went through in 24hrs.
Since the chitosan nanoparticles are very large and will not pass through any dialysis membrane, use the highest molecular weight cutoff dialysis membrane you can find. 100,000 MWCO membrane is available commercially. However, this assumes that the herbal extract will pass through. This may not be the case because the extract may be insoluble in water or it may stick to the membrane. So, the first experiment to do would be to test for the ability of the herbal extract to pass through the membrane in the absence of chitosan nanoparticles.
Also, realize that a crude herbal extract contains many substances. Some may pass through and others may not.
