I want to create a serial dilution of genomic DNA and plasmid samples for absolute qPCR. In order to get repeatable results in the Real time PCR, I want to do a very accurate dilution.
I know that I should not vortex the DNA sample because it might break, using the spinner also causes it to precipitate.
Do you think the gentle pipetting does damage the DNA? what is the best technical method for accurate diluting of DNA samples?
Note: I use "water for injection" for dilution. Are buffers (TE ,...) preferable?
