For your 1st question, I suggest that you revisit your research question again to rationalize what enzyme activity are you really after for. I suggest that you consult systematic reviews on enzyme activity/ies present in various Trichoderma spp.

Normally, enzyme assays require the addition of a substrate that the enzyme is specific for. Afterwards, you look for physical or chemical changes catalyzed by the enzyme. All this will depend on your understanding and rationalization of what enzymes to test.

I want to identify best isolates of Trichoderma and then I will use it as a biological agent against some of most important plant disease fungi such as Macrophomina phaseolina and Sclerotinia sclerotiorum. After identification of the best isolates in enzymes producing, I can test them on plant.

you can have the qunatitative estimation of enzymes and can test their antagonistic activity by dual plate test...
