For GC-MS identification of saponins it is recommended that the saponin should be hydrolysed to produce the sapogenin before analysis. However, various conditions have been recommended such as use of enzymes (glucosidases) or refluxing with 2N HCl or Sulphuric acid, in water, methanol or dioxane, for 1, 7, 22 or even 30hrs. The problem is that some of these methods produce artifacts. Can anyone who has experience with GC-MS of saponins advise on the best method/conditions to apply, as I don't have the opportunity to try many options. Secondly, what are the chances of identifying saponins with GC-MS without prior hydrolysis?
