The lipids in adipose tissue interfere with the extraction of RNA via standard Trizol method. The lipid interference is preventing the pellet formation in the Trizol method. Please suggest method by which RNA extraction can be done efficiently from adipose tissue.
use trizol method itself but a small prestep before crushing of adipose tissue can be done, use Kimwipe or strile tissue paper to press ur samples to remove excess fact and then proceed with RNA isolsation
For adipose, brain and other fatty tissues there are also some kits available that handle the high lipids. For example here are a few papers using a spin column kit for fatty tissues: http://1.usa.gov/1SDmFu1 & http://1.usa.gov/1qwIvsI
Some of these kits will recover all RNAs including microRNA as well without any TriZol.
We homogenise the tissue in TRI reagent, spin at 4oC for 10-20min at 12,000xg then pipette through the overlying fat to remove the organic layer into a fresh tube, leaving both the fat and any larger tissue debris behind. After this continue the extraction as normal. We get decent yields with this method.
