I want to purify the DNA of low copy number large plasmids from E. coli but also from non-enteric bacteria. Are there kits available for this ? What is the maximal plasmid size that may be purified with these kits and what is the yield ?
Macherey-Nagel NUCLEOBOND KIT . Use AX100 Catridges and use double volume of reactives for low copy number of plasmids. Usually , >~10 µg of plasmid DNA is amount obtained from 100 ml of E coli culture
Also the bacterial starting material is important. There is a huge difference between shaker flask cultivation and fermentation. If the plasmid is to be used for R&D, kit is ok. It is difficult to distinguish plasmid DNA from bacterial chromosomal DNA being co-purified in these systems as well.
