What is the best fluorescent indicator of Granzyme B activity for flow cytometry assays? I would prefer to label live target cells and detect active granzyme B as an indicator of cytotoxicity by CTL and NK cells.
We perform flowcytometry with primary human tissue cells/ PBMC with anibodies directed against granzyme , granzyme K and perforin (Article Characteristics of differentiated CD8(+) and CD4 (+) T cells...
). I do not know whether it is 'the best', but positivity for this granzyme B antibody corelates well with other markers of terminal differentiation on CD8+ T cells.
