What is the best bacterial suspension of 0.5 McFarland turbidity (in microlitrers) to be inoculated to 25ml of molten agar in pour plate technique to achieve semi-confluent growth?
Please post any real pictures of bacterial semi-confluent growth if you have.
A 0.5 McFarland Latex Standard is comparable to a bacterial suspension of 1.5 X 10^8 CFU/ml. 0.1 ml (100 microlitrers) of 0.5 McF works well in most bacteria.

You can use 100 microlitrers of bacterial suspension maintained at 0.5 0.5 McFarland turbidity in a Perti plate containing 25 ml molten culture media.

I purpose you to use optical density of bacterial suspension. So the best ratio is 100ul for 1mL of broth medium.

Just measure the OD and this should be good way to do the experiment. I do not think the same number applies for the different bacteria.
