How to prepare 50% DMSO from 100% DMSO?
What is the solution should I use to dilute it?
I have water and chloroform.
The plant material is initially extracted in chloroform.
Need to reconstitute a plant extract with 50% DMSO for antimicrobial activity.
If you can remove chloroform, it is preferable to use deionized water to prepare the dimethyl sulfoxide in a 1: 1 ratio,
If you cannot that, you can use chloroform to prepare dimethyl sulfoxide in a 1: 1 ratio.

You should test how chloroform-DMSO mix dilutes in water. If it works you shouldn't have a problem with both solutions. If it does not you will have to remove chloroform from your extracts and then dilute it in DMSO and finally in water. Be careful wit water dilution in the last step because to quick mixing of extract-DMSO and water could cause extract becoming insoluble. I had that happen to me when I diluted my extracts from 100% DMSO to 10%. It was I good thing that I used only a small aliquot for that mix so I haven't lost the whole sample
First the solvent of extraction should be fully removed, then the residue and the solvent of preparation should be a ratio of 1:1 as answered by Hasan Mohammed Luaibi
