I did an experiment with 6 mice using 5x105 and 10x6 cells , ( 3 mice each condition), and both cell concentration were rejected in 15 days.
I used matrigel
Cell+matrigel + 50ul
Should I use more cells? How much more?
Hello Maria, I used low passage 4T1 cells about 1x105 cells in 50 ul and I got tumors in all mice and there were no rejections at all.

Do you have wild type 4T1 cells or transduced with a viral vector? I observed an immune reaction of BalbC mice toward 4T1 cells, which resulted in rejection of primary tumour after approximately 2 weeks. However primary tumour recurred after another ~4 weeks followed by metastatic spread. I implanted 5x10^5 of 4T1 cells trancduced with vector expressing luciferase. It doesn`t happen however when you inject wild type cells. I implanted 1x10^6 and observed continuous growth of primary tumours.
