Hello everyone, may I ask what the activity assay method is for NRG1-beta 1 Protein, Human (HEK293) (HY-P73328, MCE), using regular MCF-7 cells without genetic engineering?
Seed cells at a density of 4,000 cells per well in a 96-well plate.
Remove the culture medium and add 100 μL of medium containing different concentrations of NRG1-beta 1 protein (prepared in MEM + 1% FBS + 1% P/S) to each well, so that the final concentrations of NRG1-beta 1 are 0, 0.001, 0.005, 0.01, 0.05, 0.1, 0.2, 0.5, 1, 2, 5, 10, and 50 ng/mL. Incubate for 72 h.
After incubation, under light-protected conditions, add 10 μL of CCK-8 solution to each well and incubate for 3 h at 37 °C in a 5% CO₂ incubator.
Measure absorbance at 450 nm using a microplate reader.
Note: The same method applies to testing other proteins related to MCF7 cell proliferation.
