Most people don't even consider dissolved oxygen levels in culture but its as critical as your media formulation, temperature and supplements. In open air you typically get 6.8mg/l or equivalent of 21% pO2 (really 18.5% due to humidity, but we can ignore that for now). Plenty of reports highlight the importance of O2 control for ideal cell culture outcomes, and ideally you want to culture your cells at the pO2 range experienced in vivo (3-7%).

Unfortunately, controlling pO2 in media cannot be easily accomplished, even with gas-controlled incubators and glove boxes. The only true gas control culture technology that auto-regulates the O2 level in the culture media is the Petaka G3 cell culture cassette. For best gas control, that functions automatically and self-regulates, just try the Petaka G3 and get rid of open culture dishes and flasks. Your cells will never do better.
