MIC means the concentrration in which there is no visual turbidity in the liquid medium...MBC is calculated by subculring some liquid from the concentrations showing nno turbidity, on solid media and then incubating for growth...thus MBC is always lower than MIC,,, a microbe may show no turbidity but may not be killed in the concentration...

Besides turbidity you can stain your bacterial cells (look at some of my papers) and you will see. Also you can measure on spectrophotometer the density of cells.

For both bacteria you can use Mueller Hinton Broth and micro/macro broth dilution method (CLSI M07-A09). Usually for fast growing bacteria broth methods are preferred since they are much more easier to run. 

The MIC is the lowest concentration of your drug that inhibits bacterial growth so you will have no turbidity in your culture media. But MBC is the lowest concentration that kills bacteria. Usually the concentration which is considered as MBC is higher than the concentration for MIC. On the other hand, MBC is usually presented as MBC50 or MBC90 which means the drug concentartion that kills 50% and 90%, respectively, of initial bacterial population. So, to determine the MBC you need to cultivate all of the clear tubes/wells (I mean MIC and higher concentrations) on a solid medium line nutrient agar. Then the lowest concentration of your drug that inhibits bacterial growth will be considered as MBC. 

To determine the MBC50 or MBC90, you also need to cultivate from positive control tube/well. Then the lowest concentration that inhibits the growth of 50% or 90% of bacterial colonies will be considered as MBC50 or MBC90, respectively.

MBC demonstrates the lowest level of antimicrobial agent that results in microbial death. The MBC is identified by determining the lowest concentration of antibacterial agent that reduces the viability of the initial bacterial inoculum by ≥99.9%. For bactericidal drugs, the MBC is usually the same as, and generally not more than fourfold greater than, the MIC. In contrast, the MBC of bacteriostatic drugs are many-fold greater than their MIC.

Antibacterial agents are usually evaluated as bactericidal if the MBC is less than 4 times the MIC.

In MIC you can observe the growth of bacteria by almost 80 % decline in growth

but in MBC you will not be able to observe microbial growth and to improve of that you need to culture again to find the growth

MIC is the minimum inhibitory concentration of antibiotic (drug) that prevent the bacteria to grow any more, while MBC is the minimum bacteriocidal concentration of antimicrobial which kill all bacteria.

its better to use both broth and agar, broth to see the MIC and agar is for MBC.

MIC is low concentration of antibiotics you used in Experiment can inhibit the growth of M.Os, while the MBC is the concentration of Antibiotics can kill M.Os.

In some cases may be the MIC same MBC this depend upon the bacteria and the antibiotics or whatever you use .

Regarding the calculation, you can calculate by the formula

MIC= (Lowest conc. inhibit growth + Highest conc. allow growth)/2

How long should we take to measure the MBC50 or MBC90 at a defined antibiotic concentration, 24h?

“MIC” refers to the lowest concentration of a chemical compound required to inhibit the growth of a microorganism, without necessarily killing (inactivating) the organism, after appropriate incubation period (in the case of S. aureus and E. coli it is overnight incubation at 35°C). On the contrary, “MBC” is the lowest concentration of the chemical compound required to completely inactivate (ie. required to achieve lethal effect) against the organism.

Mueller Hinton medium is preferable. Mueller Hinton medium does not have any inhibitory or antagonistic constituent. If the test chemical compound/drug is not a phytochemical, you can use Micro or Macro broth dilution technique; this technique can be used for both S. aureus and E. coli.

Based on the experience in my Lab, agar dilution method is more suitable than broth dilution when testing phytochemicals.

To calculate the “MIC”, serial dilution of the chemical agent can be made (in physiological saline, quarter-strength ringers solution, or any other physiologically suitable diluent) and incorporated into the Mueller Hinton’s broth, at known final concentrations of the drug or chemical agent; standardized inoculum density (approximately 100,000 CFU/ml) of the test organism (as well as that of control organism(s)) is, respectively, introduced into the tubes of broth. After about 20hrs of incubation (at 35°C), the lowest concentration of the drug/chemical compound that achieves inhibition of growth of the organism (in the culture medium) is the “MIC”. Subsequently, sub-cultures are made from all tubes of broth` in which there is growth inhibition (this usually includes all culture tubes with drug concentration higher or equal to that of the “MIC” tube), into tubes of fresh (sterile) Mueller Hinton broth – taking precautions to prevent “carry-over” effect of the drug to the new medium. The lowest dilution that shows absence of growth on sub-culture is the “MBC”.
