We use 6x freeze-thaw cycles and consistently show 0% viability by Trypan Blue. We have used MTT and results were good, but some say DNA assays are better.
We found out that MTT can be inhibited by Phenol Red like any other colormetric assay, but also by HOCL, which we use to oxidize NH2 terminal a.a. to make Ag uptake easier. HOCL inhibits s-glutathiaone transferase, which can affect the MTT reaction.
Our plan is to validate the MTT assay for sensitivity with serial dilution of positive control tumor cells in different wells. Then, we will do an ATP assay like Cell-titer Glo from Promega.
