I presume you want to do immunohistochemistry on mouse brain slices at E18.5? If that is the case, I recommend that you dissect the brain out very quickly and fix it in 4% PFA overnight at around 8-12 hours in the fridge. If you just submerge the brain without the dissection first the speed of fixation will be very slow and the cell viability and preservation will not be as good. It is relatively easy to open the skull and expose the brain so that the fixative can work through rapidly. Sometimes I cut sections from the dissected brain from embryonic mice to use for electrophysiology first and subsequently do the immunocytochemistry. For details see my following publications.
