I have a probe tagged with mVenus and would like to use it as a donor in a FRET experiment to detect interaction between the probe and a histone modification. I'm planning on using an antibody to detect the modification and a secondary antibody labeled with an Alexa Fluor dye to conduct FRET. I'm having difficulty finding one that has previously been published. I was thinking maybe I can use Alexa 594 but I'm not sure. I would appreciate any suggestions or personal experience.
mVenus is basically an improved version of the yellow fluorescent protein so it has a very similar spectra, just brighter. Based on the spectral shape, both Alexa 568 and 594 will work. Alexa 647 should also work but there will be less signal but also less cross talk.
