I am working on localized liver injury model in mouse to study recruitment of inflammatory immuno cell types and then specifically detecting these cell types with IHC-p. I was using F4/80 marker to detect kupffer cells but I even detected cells in untreated samples (in more or less similar frequency) giving an impression that it detects kupffer cells population in general irrespective of activation.
How to detect activated kupffer cells? Is there any marker to distinguish activated cells from non activated kupffer cells in case of IHC-p?
TNF-alpha production and high CD163 expression are considered markers for Kupffer cell activation. High plasma levels of CD163 is observed due to activation of Kupffer cells caused by portal hypertension in patients with liver cirrhosis. Try IHC with both TNF-alpha and CD163.
