Inoculate agar plugs from the margin of the colony of your culture (at least one week old) into a liquid medium (according to Dr. H. H. Ho ) and incubate it for 1 week. You can harvest their mycelia by filtration or centrifugation. Then, carefully scrape the mycelia from the agar plugs using the spatula to fully obtain the whole mycelial mass.
