I was using 10th day embryo, DMEM 450 ml, 10% FBS 50 ml, streptomycin 5 ml, tripsin for CEM culture. Three times I failed due to contamination. Finally, I got good cell growth in all the six well culture plate. But are those attachment cells myoblast cells? how can I confirm that?

Reduce your serum concentration to 2% and after 4-5 days you will be able to see myotube formation.
I hope it will work.
If you want to be sure that they are myoblasts, just check for a myoblast marker. I think pax7 should be good! Moreover primary myoblasts grow better in a medium containing chicken embryo extract.
