Hi everybody.
I have spheroids' images that I want to analyze in respect to lumen area, spheroid area and number of nuclei.
I generated a script in image J to automate my analysis, but it just works with some of the spheroids, the smaller ones and not the bigger ones. Regardless of all my attempts to adjust the filters and the thresholds, image J seems not to able to identify the lumen boundary as the signal is quite low (images attached). Now I am doing all my analysis manually which is tedious and time consuming .
My experience is quite limited to image J, so my question is there an image analysis software that can be trained to accurately identify such compartments?

One of the good image processing commercial software tools is MATLAB's image processing toolbox. It has broad ranges of built in functions and tools which are very handy and easy to use.
You can use OpenCV. OpenCV is a free open source library used in real-time image processing.
