What is a good functional assay for T cell exhaustion?

I would like to measure T cell exhaustion in an AML setting. What is a good FUNCTIONAL assay to monitor that besides phenotypic analysis (PD-1, Tim-3)?

Exhausted T cells should produce low levels of cytokines and should have low cytotoxic acitivity. Comparing the two popoulations (AML patients´s T cells vs healthy donor´s T cell) after stimulation should work.
