I am new to the use of lentiviral expression/ gene transfer vectors. From the literature I read, I gathered that lentiviral systems can be used to stably transfect mammalian cells with any gene of interest. Lentiviral systems are derived from HIV and hence have been modified in ways to generate replication deficient virions. Genes related to the synthesis of capsid proteins, polymerases, reverse transcriptases and integrases and the envelop glycoprotein are separately expressed in different plasmids, while the packaging signal is kept in a separate plasmid which also supplies the gene transfer signals.
When I looked into the exhaustive list of lentiviral plasmids available in Addgene, I wasn't sure which set of plasmids to use which will give rise to a functional virion to transfer my gene of interest into HEK293FT cells.
FInally I narrowed down to the following plasmids.
1. pWPI
2. psPAX2
3. pMD2.G
before I go to order these plasmids I want to make sure that this will do the job of stably transferring my gene of interest into HEK293.
