I am planning to isolate certain compounds from plants. Will you please give me possible factors that I should consider before proceeding to work. Thank you for your response.
1.Factors related to the plant material (parts of the plant material, collection method and time, preparation of the plant material ( drying, powdering etc)
2. The chemistry of your desired compounds (the main functional groups)
3. The physicochemical properties of your desired compounds, particularly solubility and stability
4. Selection of solvents for extraction
5. Methods of extraction
6. Preparation of crude extracts
7. Methods for purification of desired compounds (if necessary)
8. Methods for characterization of desired compounds (if necessary)
9. Methods for testing the biological/pharmacological activity test (if applicable)

Look up the plant genus and species to see what compounds have already be found so you may screen for those quickly. If doing a biological assay, see what classes of compounds tend to be active in that assay as there is a reasonable chance your active compound is in the same class (acids, bases, and so forth).
You definitely want to check the literature to see if the phytochemistry of that specific specie is known. Also, the season (time of year) that the plant material was gathered may be important as metabolites concentration can vary depending on when plant was collected.
If you are going isolate certain compounds on the basis of bioactivity then its will have some significance. There should be deep Review first. Reported basic protocols are available for fractionation on the basis of polarity. The light sensitivity, and different parameters in basic separation of molecules on the basis of material used for adoption.polarity, matrix etc.. should be consider for best achievement..
1-The analysis method should be goal-directed but should involve modern isolation and identification method (e.g. 3D HPLC, UPLC, MS etc.). These are required during publication and by relevant authorities.
2-Regarding the plant:
a-part of the plant (stolon, rhizome, root, thorn, spine, stem, leaves, flowers etc)
b-Origin: including site, altitude, the season of collection, age of harvest.
