we are try to comparing total cellular content like chlrophyll ,carotenoid ,anthocyanin, proline ,sugar ,antioxidant enzymes of medicinal plant at different altitude level of Himalayas , know about the plant response to abiotic stresses . Will it show any significant result. what else we can do to make our project more informative ?

Most of the studies related to plant response use morphological, biochemical and physiological parameters to assess changes along altitude gradient. As you have already selected important biochemical parameters in your study, I will suggest you to also assess morphological and physiological parameters if feasible. As your selected plant is medicinal, you can also assess important medicinal compounds of the plant. To further validate your data, you can measure micro-climatic factors such as meteorological and soil data at each selected altitude. Even some studies have used data of biodiversity to understand the role of altitude gradient in species distribution.

The alterations in the leaf structural elements (leaf morphological, anatomical, ultrastructural, morphometrical and photosynthetic parameters) and genome-wide investigation of molecular mechanisms for high-altitude adaptation have attracted great attention in the last few years.

There is a clear evidence that pattern of stomata distribution across the leaf and also its size coordinate well with the environmental condition. Furthermore, it has been well documented that small stomata size with high density is an adaptive response of plants to cope with dry environment. Therefore don't forget to investigate the stomatal anatomical features in your study.

My advice for you, before you start your experiment, in the begining you have to chose adequat design according to the factors that you have.
In your case I see, you have 2 Factors to accommodate
i) Plant (genotypes (n) more than one.
ii) Altitude level (n) more then 1
So, I suggest Split-plot design for this study with Altitude level as Whole-plot and genotypes as Sub-plot
the most important thinks you have to set the same experimental design in different altitude level like this you analyse your data.

Morphological and phenological studies are to be carried out . Any species or for that matter any living thing will try to adjust by modifying itself to adjust to the new environment and will try to reproduce leading to a new generation with greater adaptability.
Try to make analysis on the plant root fungi. Because mostly they play key roles.
