I'm trying measuring the concentration of protein in solution. I know what kind of protein is, but protein concentration and buffer condition are not known.

I want to know its concentration approximatively. So, I checked the UV absorbance value (A280) using Nanodrop. There was a peak at A280 but also at A235 - value was lowered when dilute for 1:10 and 1:100. I think buffer composition influence absorbance at 230nm, but the peak at A235 was too high.

I read secondary and tertiary structure of protein have absorbance at 235nm but I'm not sure the calculation using only A280 value (and estimate extinction coefficient) is meaningful.

What dose A235 value mean for protein solution?
