Is it correct that a hybridoma has the full intact genome of both the splenocyte and the myeloma to which it is fused? Or do genomic reorganizations take place post fusion?
What about the genomic region in the splenocyte which harbors the immunoglobulin genes, do these remain unaltered during the entire immortalization process?
The myeloma line is often tetraploid and the spleen cell diploid; the fusion results in a mixed population not purely hexaploid, but a range from hypertetraploid, hyperpentaploid and hexaploid. Naturally cloning will select those cells that will stably produce the desired antibody, but nevertheless stability of something not proven by natural selection over time is a koan.
The heterokaryon (synkaryon) packaging all of the chromosomes was a reason why mouse fusion lines were used instead of pure human, which tended to form multikaryons. The link below recites tracking of lines over two years, mentioning that chromosomes can tend to be lost after fusion, and that dexamethasone has an effect on maintaining ploidy level.
I don't think you can assume the full genome of both, nor would I expect over long periods and under the stress of continuous replication that a line will be completely stable.
