or you missed a copy number possibility. do you know that the genome bears about 20-25k genes and that P53 is not the only one causing cancers? I invite you to take a look at the TCGA database (https://www.cancer.gov/about-nci/organization/ccg/research/structural-genomics/tcga) to get other targets.

qPCR tells you expression levels and/or copy level, not whether there are mutations present in the region being amplified.
